How VapoFil Works: The Three Mechanisms Claimed For This Blend

Three stories are told about blends like this one: hormone binding through nettle lignans, nitric oxide through icariin, and adaptogenic support through tongkat ali. All three are real chemistry. Only one has human trials behind it, and those trials used ten to forty times what this panel prints.

Mechanism and effect are different claims. A mechanism explains why an ingredient was chosen. Only a trial shows what happens when a person takes it.

The hormone story

Mechanism one: how VapoFil is said to free up testosterone

Most testosterone in circulation is bound to two proteins, and the largest share is carried by sex hormone binding globulin. The proposal is straightforward: occupy some of that binding capacity with something else, and more testosterone stays unbound and available.

The laboratory work behind it isolated lignans from nettle and their metabolites, and showed that they bind to human SHBG in an assay. That is a genuine finding and it is where the whole story comes from.

Two things get lost between that assay and a capsule. The lignans came from nettle root, and this panel prints nettle leaf. And binding in a test tube at whatever concentration the experiment used is not the same as 20 mg of a leaf extract changing free testosterone in a man. No trial has measured that.

Why the root and leaf distinction is not pedantry

The twelve-month trial in this area, Schneider and Rübben's, used root at 459 mg a day. The review of the evidence is titled for the root, urticae radix, because that is the part that was studied. Leaf has been trialled for other things entirely — blood sugar in type 2 diabetes is the controlled study that exists.

The blood-flow story

Mechanism two: how VapoFil is said to help circulation

An erection is a blood-flow event. Nitric oxide relaxes the smooth muscle in the arteries that supply the penis, an enzyme called PDE5 breaks down the signal that nitric oxide starts, and prescription erectile medicines work by inhibiting that enzyme so the signal lasts.

Icariin, the marker compound in horny goat weed, inhibits the same enzyme. That was measured on isolated human PDE5 and it is the reason this plant is in so many products. Later work on synthetic analogues mapped which parts of the molecule drive the effect, and found modifications hundreds of times more potent than icariin itself.

Which is the useful clue. Icariin is a weak inhibitor by the standards of the drugs it is compared to, the rat work showing improved erectile function used amounts scaled to a rat, and there is no randomised trial in men. This panel prints 8 mg of a 10:1 extract, stated as 80 mg of dry herb.

The adaptogen

Mechanism three: the adaptogen on the VapoFil panel

Tongkat ali is the ingredient with a real human file behind it. A 2022 systematic review and meta-analysis pooled the clinical trials and found an increase in serum total testosterone across them. Individual studies have reported improvements in quality-of-life and sexual well-being scores, in a 2012 trial and in a 2014 trial of the same extract with Polygonum minus, and a six-month 2021 trial looked at erectile function in men with age-related androgen decline.

Every one of those used a standardised aqueous root extract at 100 to 400 mg a day. The 2021 multicentre study is typical. This panel prints 10 mg of a 100:1 concentrate, stated as equivalent to 1,000 mg of dry powder, which is a different measure from a standardised extract dose and cannot be read across to it.

The distinction this page turns on
  • A 100:1 ratio describes the input. A standardisation describes the output.
  • A trial dose belongs to the preparation the trial used, not to the plant.
  • Two extracts at the same ratio can differ several-fold in active content, which is the whole of the standardisation problem.
  • None of that means 10 mg does nothing. It means nobody has measured what it does.
The rows with no story

What the VapoFil panel does not have a mechanism for

Three rows carry no mechanism worth printing. Wild yam's diosgenin cannot be turned into hormones by a human body, and the only controlled trial found no hormonal change over three months. Sarsaparilla has chemistry in the steroidal saponins the root carries and no clinical trial at all. Boron at 1 mg of elemental boron is an ordinary day's dietary intake, which reviews of boron in nutrition describe as the amount most people already get.

Saw palmetto is the fourth and most interesting case. It has plenty of mechanism and a very large evidence base, and the 2023 Cochrane review found that at the researched dose it does little to nothing for the symptoms it is sold for. Being well studied and being effective are separate questions.
